New cancer 'vaccine' shows future promise in treating and preventing metastatic cancers

Preclinical, laboratory studies suggest a novel immunotherapy could potentially work like a vaccine against metastatic cancers, according to scientists at Virginia Commonwealth University Massey Cancer Center. Results from a recent study show the therapy could treat metastatic cancers and be used in combination with current cancer therapies while helping to prevent the development of new metastatic tumors and train specialized immune system cells to guard against cancer relapse.

Recently published in the journal Cancer Research, the study detailed the effects of a molecule engineered by lead author Xiang-Yang Wang, Ph.D., on animal and cell models of melanoma, prostate and colon tumors. The molecule called Flagrp-170 consists of two distinct proteins, glucose-regulated protein 170 (Grp170), known as a "molecular chaperone," and a "danger signal" derived from flagellin, a protein commonly found in bacteria. The researchers used modified viruses, or adenoviruses, that can no longer replicate to transport Flagrp-170 directly to the tumor site to achieve localized vaccination. The novel therapy caused a profound immune response that significantly prolonged survival in animal models.

"Successfully promoting antitumor immunity will help eradicate tumor cells, control cancer progression and help prevent tumor relapse," says Wang, Harrison Scholar, member of the Cancer Molecular Genetics research program at VCU Massey Cancer Center and associate professor of Human and Molecular Genetics at VCU School of Medicine. "This immunotherapy has the potential to be used alone or in combination with conventional cancer treatments to develop and establish immune protection against cancer and its metastases."

Grp170 is currently being explored for its potential as a "cancer vaccine" because it has been shown to help the immune system recognize cancer antigens. Antigens are molecules from foreign objects such as bacteria, viruses or cancer that, when detected, provoke an immune response aimed at attacking them. However, because cancer cells can alter the microenvironment surrounding a tumor, they are able to suppress immune responses and continue replicating without being attacked by the body's natural defenses.

The chimeric chaperone Flagrp-170, created by strategically fusing a fragment of flagellin to Grp170, not only enhances antigen presentation, it also stimulates additional immune signals essential for functional activation of specialized immune cells, including dendritic cells, CD8+ T lymphocytes and natural killer (NK) cells. Dendritic cells act as messengers between the innate and adaptive immune systems. Once activated in response to a stimulus such as Flagrp-170, dendritic cells migrate to lymph nodes where they interact with other immune cells such as T lymphocytes to shape the body's immune response. CD8+ T lymphocytes and NK cells are known to respond to tumor formation and kill cancer cells by triggering apoptosis, a form of cell suicide.

"Overcoming cancer's ability to suppress the body's natural immune responses and restore or develop immunity for tumor eradication is the goal of cancer immunotherapy," says Wang. "More experiments are needed, but we are hoping Flagrp-170 may one day be used in formulating more effective therapeutic cancer vaccines."

Moving forward, Wang and his team are working to better understand the molecular mechanisms responsible for Flagrp-170's therapeutic effects. Additional studies are underway to more efficiently target and deliver Flagrp-170 to tumor sites in order to provoke a more robust and durable immune response.

Amazon corrals FX's Justified to exclusive Prime Instant Video deal

If Amazon's recent foray into British period drama leaves you cold, its Prime Instant Video service now has something grittier on tap: The FX Network's Justified. Thanks to a new deal with Sony Pictures Television, you can download the western-style crime series for no additional cost if you're a subscriber on the $79 yearly service. It was up for grabs at $1.99 a pop on Instant Video before, where Amazon said it was one of its most popular buys, and now Prime subscribers can grab episodes from seasons one through three. HBT: Minor leaguer talks about PED pressure

While most of you like to call me a steroids apologist, the fact of the matter is that I am not pro-steroids. I am anti-hysteria and distortion. I am also against a baseball writing community that overwhelmingly thinks that the most worrisome and therefore most newsworthy aspect of PEDs in baseball is what it means for the record books and the Hall of Fame when there are far more important implications of PED use.

If you don?t believe me, allow me to quote myself from April 2007 ? the very month I began blogging about baseball on a regular basis. It was around that time ? months before the Mitchell Report came out ? that Kirk Radomski was making news and the names of some marginal players to whom he dealt were coming out. ?I opined then that, once we know more about PEDs in baseball, we?ll see that it?s likely a bigger problem among those marginal players ? the guys trying to crack the bigs or hang on; the 26th man in the organization who feels he need that extra oomph ? than it is among superstars:

I don?t say this in an effort to minimize the steroid problem. Indeed, minor?leaguers?and players who aren?t superstars constitute the vast majority of professional ballplayers. If my theory holds, the problem could be far greater than that which is portrayed by sportswriters who like to caricature only the most prolific sluggers as juicers. If I?m right, our concern over records and the Hall of Fame would seem pretty petty in comparison to the scores of regular?Joes?who are ruining their health as they walk the line between a lifetime of comfort and a?job at a warehouse. Players that the steroid moralizers in the media almost uniformly ignore.

Now, I got a few things wrong back then, of course. I probably underestimated the number of superstars who used PEDs and I hilariously lumped Alex Rodriguez in with the non-users because that?s the best information anyone had back then. But I think the dynamic still holds: it?s a way, way bigger moral problem for a marginal player to feel like he has no choice but to take steroids than it is ?for an already great baseball player to feel like he should take steroids to break some records.

This doesn?t mean that the superstars aren?t cheaters if they take PEDs and it doesn?t mean that they shouldn?t be held to account. What I?m getting at is that, in the great baseball conversation about PEDs, we should not care nearly as much as we do about records and legacies and we should care far more about what PEDs are doing down at the lower levels of baseball. ?We should spill way less ink about who we think ?the real Home Run King? is ? as if that matters ? and think way harder about those frequent minor league suspensions and what they mean to the people who are faced with the choice to take dangerous drugs or wind up out of baseball.

Against that backdrop is this excellent column from Eric Knott. Knott pitched 11 years in the minors and 24 games in the majors. He is the quintessential borderline guy who, if he had an extra couple of miles per hour on his heater, may have stuck. ?But he didn?t get those miles per hour, and he didn?t try PEDs in an effort to do so.

Knott gives a fascinating, clear-eyed and detailed rundown of the environment in baseball during the height of the Steroid Era, as well as what factored into his decisions about whether to use.

It?s an absolute must-read. There?s more useful information in this piece than anything that can be found in the Mitchell Report or the latest bombastic anti-PEDs screen from Johnny Sportswriter.

FDA expands approval of Bayer cancer drug

WASHINGTON (AP) -- The Food and Drug Administration on Monday expanded approval of a Bayer cancer pill to treat tumors of the intestinal tract that don't respond to other treatments.

The drug is called Stivarga and regulators approved it to treat gastrointestinal stromal tumors that cannot be surgically removed and no longer respond to other FDA-approved drugs.

The FDA previously approved Stivarga to treat colorectal cancer. It works by blocking several enzymes that promote cancer growth.

The FDA approved the drug for the new use based on a study of nearly 200 patients who were randomly assigned to take Stivarga or a placebo pill.

Patients taking the drug experienced a nearly four month delay in the growth of their tumors compared to taking placebo.

Serious side effects of Stivarga in clinical trials included liver damage, severe bleeding, blistering and peeling of skin, high blood pressure, heart attacks and perforations.

Other drugs approved to treat intestinal tumors include Gleevec, from Novartis, and Sutent, made by Pfizer Inc.

Cleric resigns after allegations of 'inappropriate' conduct with priests

LONDON (Reuters) - Britain's most senior Roman Catholic cleric resigned on Monday following allegations he behaved in an inappropriate way with priests, and said he would not take part in the election of Pope Benedict's replacement.

Cardinal Keith O'Brien said he had tendered his resignation some months ago, ahead of his 75th birthday in March and because he was suffering from "indifferent health".

The Vatican said the pope, who steps down on Thursday, had accepted O'Brien's resignation as archbishop of St. Andrews and Edinburgh.

O'Brien, an outspoken opponent of gay marriage, has been reported to the Vatican over allegations of inappropriate behavior stretching back 30 years, according to Britain's Observer newspaper.

The cardinal, who last week advocated allowing Catholic priests to marry as many found it difficult to cope with celibacy, rejected the allegations and was seeking legal advice, his spokesman said.1

"Looking back over my years of ministry: For any good I have been able to do, I thank God. For any failures, I apologize to all whom I have offended," O'Brien said in a statement, which made no reference to the recent allegations.

He said he would not attend the election next month of a new pope, saying: "I do not wish media attention in Rome to be focused on me - but rather on Pope Benedict XVI and on his successor."

The Observer, which gave little detail on the claims, said three priests and a former priest, from a Scottish diocese, had complained over incidents dating back to 1980.

One said the cardinal formed an "inappropriate relationship" with him while another complained of unwanted behavior by O'Brien after a late-night drinking session.

Last year, O'Brien's comments labeling gay marriage a "grotesque subversion" landed him with a "Bigot of the Year" award from British gay rights group Stonewall.

Crews looks for 4 in waters south of San Francisco

HALF MOON BAY, Calif. (AP) ? The Coast Guard was searching the rough waters south of San Francisco late Sunday for four people, including two children under 8, who went missing after they reported their sailboat was sinking.

The group was approximately 65 miles off Pillar Point Harbor in Half Moon Bay around 4:20 p.m. Sunday when they made a distress call, Lt. Heather Lampert said.

The group said their 29-foot sailboat was taking on water and their electronics were failing. The bay about 25 miles south of San Francisco.

An hour later, the group reported that it was forced to abandon their boat. They didn't have life rafts on board, Lampert said, and were trying to make one out of a cooler and life preserver ring.

Lampert said the Coast Guard then lost radio contact with the group.

The National Weather Service had issued an advisory throughout the weekend warning boaters of strong winds and rough seas around the Bay Area.

Mariners "operating smaller vessels should avoid navigating in these conditions," the advisory said.

Lampert said Coast Guard crews have conducted several searches using a fleet of boats, helicopters and airplanes, but have not yet found signs of the group.

More searchers from the California Air National Guard, and Coast Guardsmen aboard a 210-foot cutter, were expected to join the search through the night.

Half Moon Bay is about 25 miles south of San Francisco.

ANAHEIM, Calif. -- Robbie Lawler came back to the UFC with a big win as he knocked out Josh Koscheck on Saturday at UFC 157.

NRA uses Justice memo to accuse Obama on guns

WASHINGTON (AP) ? The National Rifle Association is using a Justice Department memo it obtained to argue in ads that the Obama administration believes its gun control plans won't work unless the government seizes firearms and requires national gun registration ? ideas the White House has not proposed and does not support.

The NRA's assertion and its obtaining of the memo in the first place underscore the no-holds-barred battle under way as Washington's fight over gun restrictions heats up.

The memo, under the name of one of the Justice Department's leading crime researchers, critiques the effectiveness of gun control proposals, including some of President Barack Obama's. A Justice Department official called the memo an unfinished review of gun violence research and said it does not represent administration policy.

The memo says requiring background checks for more gun purchases could help, but also could lead to more illicit weapons sales. It says banning assault weapons and high capacity ammunition magazines produced in the future but exempting those already owned by the public, as Obama has proposed, would have limited impact because people now own so many of those items.

It also says that even total elimination of assault weapons would have little overall effect on gun killings because assault weapons account for a limited proportion of those crimes.

The nine-page document says the success of universal background checks would depend in part on "requiring gun registration," and says gun buybacks would not be effective "unless massive and coupled with a ban."

The administration has not proposed gun registration, buybacks or banning all firearms. But gun registration and ownership curbs are hot-button issues for the NRA and other gun-rights groups, which strenuously oppose the ideas.

Justice Department and White House officials declined to provide much information about the memo or answer questions about it on the record.

The memo has the look of a preliminary document and calls itself "a cursory summary" and assessment of gun curb initiatives. The administration has not release it officially.

But the NRA has posted the memo on one of its websites and cites it in advertising aimed at whipping up opposition to Obama's efforts to contain gun violence. The ad says the paper shows that the administration "believes that a gun ban will not work without mandatory gun confiscation" and thinks universal background checks "won't work without requiring national gun registration" ? ideas the president has not proposed or expressed support for.

"Still think President Obama's proposals sound reasonable?" Chris W. Cox, the NRA's chief Washington lobbyist, says in the ad.

Last month, White House spokesman Jay Carney said none of Obama's proposals "would take away a gun from a single law-abiding American." Other administration officials have said their plans would not result in gun seizures or a national gun registry.

A Justice Department official who would only discuss the issue on condition of anonymity said the NRA ad misrepresents Obama's gun proposals and that the administration has never backed a gun registry or gun confiscation.

While the memo's analysis of gun curb proposals presents no new findings, it is unusual for a federal agency document to surface that raises questions about a president's plans during debate on a high-profile issue such as restricting firearms.

Obama wants to ban assault weapons and ammunition magazines exceeding 10 rounds that are produced in the future. He wants universal background checks for nearly all gun purchases. Today, checks are only mandatory on sales by federally licensed gun dealers, not transactions at gun shows or other private sales.

His plan also includes tougher federal laws against gun trafficking and straw purchases, which occur when a person legally buys a firearm but sells it to a criminal or someone else barred from owning a weapon.

Interest in the gun issue has intensified since the December shootings in Newtown, Conn., that killed 20 first-graders and six staffers at an elementary school. The Democratic-led Senate Judiciary Committee plans to write legislation addressing some of Obama's proposals in the next week or two.

The NRA's Cox declined to say how his organization obtained the memo.

He said the commercial is running online in 15 states, including many Republican-leaning states where Democrats will defend Senate seats next year, such as Alaska, Arkansas, Louisiana, Montana, North Carolina, South Dakota and West Virginia. There are also ads in papers in five states.

The memo was written under the name of Greg Ridgeway, acting director of the National Institute of Justice, the Justice Department's research arm. It is dated Jan. 4, nearly two weeks before Obama announced his plan for restricting guns, and Ridgeway's first day as acting chief.

Justice Department officials said Ridgeway was not granting interviews. He came to the institute last July from the RAND Corporation, a nonprofit research institution where he studied criminal justice issues, and has a Ph.D. in statistics.

The memo says straw purchases and gun thefts are the largest sources of firearms used in crimes, and that such transactions "would most likely become larger if background checks at gun shows and private sellers were addressed."

Gun control supporters said the NRA ad and the Justice memo don't mention that the current federal background check system blocked gun sales to 2.1 million criminals and others barred from owning guns between 1994, when the checks began, and 2010. Also ignored is that Obama has proposed cracking down on straw purchases to prevent a growth in illegal transactions, they said.

Advocates of restricting guns also said the memo omitted mention of several studies that affirm the effectiveness of firearms curbs. These include a 2010 police group analysis showing more than one-third of police departments found increased criminal use of assault weapons and high-capacity magazines since the 2004 expiration of the ban on those items.

"It doesn't appear to be a serious discussion of gun violence prevention policy, never mind an expression of administration policy," said Joshua Horwitz, executive director of the Coalition to Stop Gun Violence.

The memo says that out of 11,000 annual gun homicides, an average of 35 deaths yearly are from mass shootings, defined as those with four or more victims.

"Policies that address the larger firearm homicide issue will have a far greater impact even if they do not address the particular issues of mass shootings," it says.

It says there were an estimated 1.5 million assault weapons before the 10-year ban on those firearms began in 1994, so their sheer number would weaken a new ban exempting existing weapons. Such guns accounted for just 2 percent to 8 percent of crimes before the 1994 ban, so eliminating assault weapons "would not have a large impact on gun homicides," the memo said.

Recent data on the assault weapons ban impact is scarce because since the 1990s, Congress has blocked most federal research on the effect that firearms have on public health. As part of the gun restrictions Obama proposed last month, he ordered federal scientific agencies to research gun violence.

CAIRO (AP) ? A key opposition leader called Saturday for a boycott of upcoming parliamentary elections, saying he will not take part in a "sham democracy."

President Mohammed Morsi's Islamist party, the Muslim Brotherhood, shot back that the opposition was running away from the challenge and wants power without contesting elections.

The exchange reflected a new escalation in political tensions that could spill into even wider strikes and protests ahead of a four-stage vote set to begin on April 28 and last until June. Morsi announced the elections late Thursday night.

"(I) called for parliamentary election boycott in 2010 to expose sham democracy. Today I repeat my call, will not be part of an act of deception," Nobel laureate Mohammed ElBaradei, who leads the main opposition National Salvation Front, wrote on his Twitter account.

He reiterated the opposition's refrain that Morsi, who was elected in a free and fair vote, is acting like former autocratic leader Hosni Mubarak.

Almost immediately after ElBaradei's call, rifts began appearing in the opposition which has struggled to unite since it led the successful uprising two years ago that ousted Mubarak.

Some activists criticized the boycott call, saying it would alienate the masses and allow the Islamists to maintain their domination of parliament.

The Brotherhood has emerged from the uprising as Egypt's most powerful political group, winning both parliamentary and presidential elections.

The deputy head of the Brotherhood's Freedom and Justice Party, Essam el-Erian, responded to ElBaradei's call on his Facebook page saying "running away from a popular test only means that some want to assume executive authority without a democratic mandate."

"We've never yet known them to face any election or serious test," he said of the opposition leaders.

In Egypt's first free elections in 2011, the Brotherhood won nearly half of seats in parliament and the more conservative Salafis won a quarter, making the two Islamist groups dominant.

Liberal and secular parties have trailed significantly in all elections since Mubarak was toppled. Their outreach across Egypt, while growing, is still dwarfed by the Islamists' well organized network of charities and programs that assist the poor.

Nearly half of Egypt's 85 million people live below the poverty line, subsisting on less than $2 per day.

Blogger and commentator Mahmoud Salem, a longtime activist who protested against Mubarak and now opposes Morsi, said he disagreed with boycott calls because it offers no real alternative to the current political impasse.

"Where's ElBaradei's party, its plan, its economic vision? Let's say a boycott is the right answer. What will they do so that they can be competitive in the next election?" Salem said.

He said that ElBaradei is also partly calling for a boycott of the vote because the opposition has been unable to win a significant number of seats.

"In reality, it will end up as a parliament composed of the Muslim Brotherhood, the Salafis, or members of the ex-regime," he said.

The opposition has accused Morsi and his Brotherhood backers of using election wins to monopolize power in tactics similar to the former regime. They accuse him of reneging on a promise to form an inclusive government representative of the Christian minority, women, and liberals.

Elections under Mubarak's three-decade rule were widely rigged and the chamber was dominated by members of his ruling party.

The state-run MENA news agency reported that the president is studying changing the starting of date of elections following an outcry by Coptic Christians in Egypt.

The first phase coincides with Palm Sunday and Easter for Egypt's minority Christians, who tend to travel during the holidays and have consistently voted against the Muslim Brotherhood.

ElBaradei's opposition coalition, which was only formed late last year, had warned for weeks it could boycott parliamentary elections if certain conditions were not met first.

The NSF said it wants a real national dialogue that leads to the formation of a national unity government, changes to the new constitution and stability.

On the second anniversary of the Jan. 25 uprising this year, anger spilled out onto the streets and violence again engulfed the nation.

About 70 people died in a wave of protests, clashes and riots in the past four weeks, and more than half were killed in the city of Suez Canal city of Port Said alone.

The lower house of parliament, which drafts laws in Egypt, was disbanded on June 14, 2012 after the Supreme Constitutional Court ruled that a third of the chamber's members were elected illegally. The upcoming elections are aimed at reinstating the legislature.

Former lawmaker Mostafa al-Naggar, a centrist who beat an ultraconservative, popular Salafi candidate in 2011, said calls for a boycott will be ineffective unless there is unity among the opposition. He said mixed messages will confuse voters and push people away.

He wrote on Twitter that "the decision to boycott the next elections is extremely dangerous because it will clear the arena for the ruling party and its allies to dominate the legislative and executive authorities."

Morsi's supporters say that delaying elections, protesting and boycotting after years of autocracy under Mubarak is affecting Egypt's ability to lure foreign investors and tourists again.

The political unrest has hit Egypt's foreign currency reserves, which have fallen below a critical level to less than $14 billion.

Meanwhile, many residents of the city of Port Said blame Morsi's policies for the turmoil.

A civil disobedience campaign in the city started a week ago.

More than 1,000 people, including hundreds of employees of the Suez Canal Authority, protested Saturday outside one of the vital waterway administration's gates. Shipping in the international waterway has not been affected.

The protesters are demanding retribution for those killed during unrest in the city and for officials to be on trial. On Friday, around 15,000 protested against Morsi and hung effigies of him in the main square there.

There have also been near daily protests in Cairo and in the textile producing city of Mahalla.

Since Morsi's election win as Egypt's first civilian and Islamist president last summer, his popularity has eroded.

Thousands took to the streets in December when he issued power-grabbing decrees temporarily that allowed his supporters to rush a draft constitution to a nationwide vote before a high court packed with Mubarak appointees could disband the process.

It passed with 64 percent amid low turnout and a boycott by thousands of overseeing judges.

Boeing gives regulators its plan to fix Dreamliner batteries

WASHINGTON/SEATTLE (Reuters) - Boeing Co on Friday gave U.S. aviation regulators its plan to fix the volatile battery aboard its new 787 Dreamliner, even though investigators have not yet determined what caused the batteries to overheat on two planes last month.

Boeing did not propose abandoning the lithium-ion batteries and is not working on a backup or longer-term fix for the problem that has grounded its entire fleet of 50 Dreamliners for nearly five weeks, three sources familiar with the plan said.

The company and the U.S. Federal Aviation Administration said no firm result emerged from the meeting between Deputy Transportation Secretary John Porcari, FAA Administrator Michael Huerta and other FAA officials and Boeing Commercial Airplanes CEO Ray Conner and other senior Boeing executives in Washington.

With Boeing's costs mounting by millions of dollars a day while the planes are on the ground, the FAA said it is "reviewing a Boeing proposal and will analyze it closely. The safety of the flying public is our top priority and we won't allow the 787 to return to commercial service until we're confident that any proposed solution has addressed the battery failure risks."

Boeing declined to comment on the details of its proposal, but said the meeting with the FAA was productive.

The proposal to the FAA includes measures to address a range of possible causes of short-circuits in the batteries, the sources said.

Five weeks ago, U.S. authorities grounded the worldwide fleet of 787s. U.S., Japanese and French investigators are still not certain what caused the battery fire aboard an All Nippon Airways 787 in Boston and an overheated, smoking battery on a Japan Airlines 787 in Japan.

The proposed fix includes adding ceramic insulation between the cells of the battery to help keep cells cool and prevent a "thermal runaway" in which one cell overheats and triggers overheating in adjacent cells. It also includes building a stronger, larger stainless steel box with a venting tube to contain a fire and expel fumes outside the aircraft should a battery catch fire again, the sources said. In addition, the plan proposed wiring changes, self-torquing screws that will not come loose and battery alterations to prevent moisture and vibration problems, one of the sources said.

But there was also a plan to use a different battery type or some other longer-term fix, the sources said.

"I have talked to a number of people who are working directly on these batteries. No one is on the Plan-B team," said a person familiar with Boeing's efforts who was not authorized to speak publicly about them.

A second source, who also was not authorized to speak publicly, said Boeing does not view its proposal as a temporary "band-aid" that would be supplanted by another solution later.

Boeing spokesman Marc Birtel said in a statement: "We are encouraged by the progress being made toward resolving the issue and returning the 787 to flight for our customers and their passengers around the world."

Birtel reiterated that hundreds of engineers and technical experts are working "around the clock" to return the 787 fleet to service. "Everyone is working to get to the answer as quickly as possible and good progress is being made," Birtel said.

Boeing's stock closed up 65 cents, or 0.86 percent, at $75.66 on the New York Stock Exchange.

Richard Aboulafia, aerospace analyst with the Teal Group in Virginia, said Boeing needed a backup plan in case the FAA did not approve its proposal.

"It's a bit tone deaf to propose containment and management when the political winds are favoring an elimination of the risk," he said, citing Transportation Secretary Ray LaHood's insistence that the plane would return to flight only when it was "1,000 percent safe" and similar remarks by other officials.

"They need to be out there talking about a bigger solution beyond mere containment because the political winds and public opinion are not going to favor a solution that's focused on fire and smoke management," Aboulafia said.

He noted that Airbus had already signaled its plan to switch back to more traditional nickel cadmium batteries for its A350 airliner, but the 787 was far more dependent on electrical power, which would complicate any effort to switch to a different type of battery. A complete redesign could take around nine months to implement, he said.

Another source said that kind of solution could take two years if, for example, Boeing decided to use nickel cadmium batteries on the 787, similar to those used on the 777 jet.

The U.S. National Transportation Safety Board is still investigating the Boston fire and the Japan Transportation Safety Board is investigating the battery failure in Japan. Neither has found a root cause for the problems.

The sources said the NTSB might never find the cause because the battery in Boston was severely damaged by the fire.

Given the financial cost of the grounding for Boeing and the airlines that own the jets, estimated at $200 million a month, Boeing decided to address all possible causes with the measures, rather than wait for the NTSB to identify one specific cause, the sources said.

Boeing engineers have been working with outside experts and U.S. government officials to address possible cause of the battery issues. The team includes experts from the U.S. Navy and the National Aeronautics and Space Administration, which uses a lithium-ion battery on board the International Space Station.

Boeing engineers went through a "fault tree" and "came up with a list of half a dozen things that could have led to problems," said a congressional source who had been briefed on the matter, but was not authorized to speak publicly.

"They have a list of things that it could be, and the fixes are designed to address that list of problems," the source said.

If the NTSB's investigation turns up additional possible causes, those would be added to the mix, another of the sources said.

Boeing machinists already are building the new containment boxes for the battery, a sign the company is confident that the FAA will eventually approve continued used of lithium batteries and the contain-and-vent strategy for dealing with fires, according to a person familiar with the matter.

The FAA granted Boeing permission to use lithium batteries in 2007, but set nine special conditions the company had to meet.

Asked why the company's extensive testing of the batteries had not revealed problems with the batteries and the electrical systems used to operate them, one of the sources said test environments had limitations and the real test of an aircraft always came when it was actually operating.

If the Boeing plan is approved by FAA Administrator Huerta and Transportation Secretary LaHood, company officials expect the 787 fleet to return to service within eight weeks, one source said.

Another source, who is also familiar with the 787 investigation but not authorized to speak publicly, said a key challenge for Boeing would be to redesign the battery box so that it could truly contain a fire if one occurred.

Despite Boeing's statements about containment being the plan for a battery issue from the start, the blue box that held the current lithium-ion battery was clearly "not designed to contain a fire," said the source.

Another person familiar with the engineering work said the new box would be made of stainless steel nearly half an inch thick. It would be capable of containing an explosion, and would have a tube to vent smoke and flame outside the jet.

However, the source said engineers have raised questions about the safety of venting flames outside the plane, especially if it is on the ground and being fueled. The effect could be something like a flamethrower, this person said.

The new state funding would ensure financial stability for the New Mexico Bowl, while purchasing a significant amount of TV airtime for New Mexico Tourism Department commercials during the game?s national broadcast on ESPN, Rep. Antonio ?Moe? Maestas, D-Albuquerque, told the Journal.

?To have a bowl of this magnitude touting New Mexico in our largest city, I think, is a win-win,? Maestas told the Journal. ?? The economic impact is clear to the city of Albuquerque, and the advertising for the entire state bolsters the entire state tourism industry.?

The effort to increase the state funding for the game is co-sponsored by Rep. Nate Gentry, R-Albuquerque, the Journal reported.

An estimated 24,610 fans attended the 2012 Gildan New Mexico Bowl to see Arizona beat Nevada 49-48. That attendance fell far below the game?s previous attendance record of 34,111 set during the inaugural game in 2006, when the University of New Mexico lost 20-12 to San Jose State.

Tourism Department Secretary Monique Jacobson, prior to the 2012 game, said her office hadn?t see a significant return on its investment in the game beyond the $50,000 cost of keeping New Mexico in the bowl title, according to the Journal report.

Obama challenges GOP on economy in State of the Union

WASHINGTON (CNN) -

President Barack Obama challenged Congress to join him in taking on "our generation's task" to ignite the growth of a "rising, thriving middle class," using the first State of the Union speech of his second term to prod Republicans to compromise on the major challenges facing the nation.

"It is our unfinished task to restore the basic bargain that built this country -- the idea that if you work hard and meet your responsibilities, you can get ahead, no matter where you come from, what you look like, or who you love," Obama said Tuesday night, sounding familiar themes from his re-election campaign last year.

The president emphasized economic growth and job creation, and insisted that his proposals would not increase the nation's federal deficit.

He also made an emotional plea for Congress to hold votes on controversial proposals for tougher gun laws after the Newtown, Connecticut, shootings in December that killed 20 schoolchildren.

At the same time, Obama called for legislators to work together for the good of the nation, saying Americans "expect us to put the nation's interests before party."

"They do expect us to forge reasonable compromise where we can," he said. "For they know that America moves forward only when we do so together, and that the responsibility of improving this union remains the task of us all."

It was his fourth State of the Union address and seventh speech to a joint sitting of Congress, and analysts considered it a crucial moment for setting the tone for the political dialogue after four years of partisan division and congressional dysfunction.

With the government facing deep spending cuts mandated by a previous agreement between Obama and Congress to raise the federal debt ceiling, Obama renewed his call for a comprehensive deficit-reduction plan that includes new tax revenue coupled with spending cuts.

Taking aim at the bitter partisanship of his first term, Obama said "let's set party interests aside, and work to pass a budget that replaces reckless cuts with smart savings and wise investments in our future."

"And let's do it without the brinksmanship that stresses consumers and scares off investors," he continued to applause, mainly from Democrats. "The greatest nation on Earth cannot keep conducting its business by drifting from one manufactured crisis to the next. We can't do it. Let's agree, right here, right now, to keep the people's government open, and pay our bills on time, and always uphold the full faith and credit of the United States of America."

In a jab at congressional Republicans who seek to shrink deficits and the size of government through deep spending cuts, saying "deficit reduction alone is not an economic plan."

However, a hard-line stance by Obama on deficit reduction after forcing Republicans to concede already on tax increases could poison prospects for the comprehensive deal he says he wants, CNN Senior Political Analyst David Gergen noted.

"The window for achieving a grand bargain has been closing rapidly and could slam shut Tuesday night," Gergen wrote.

After arriving a loud cheers and prolonged applause on a night of political pomp and ceremony, Obama began the speech on a positive note, saying the nation was on sound footing to move forward.

"Together, we have cleared away the rubble of crisis, and can say with renewed confidence that the state of our union is stronger," he said.

Obama's address comes amid breaking news around the world that touched on the challenges facing his government and the nation.

A suspected North Korean nuclear test and a gun battle involving an alleged cop killer dominated headlines in the hours before the speech.

Meanwhile, the Dow Jones Industrial average closed at its highest level since 2007, exceeding the 14,000 mark, even though unemployment continued to hover near 8%.

Obama continue his push for Congress to act on politically volatile issues such as immigration reform. Other measures in the speech included a paycheck fairness act intended to make it easier for women to fight salary discrimination without losing their jobs, and new proposals to develop alternative energy hubs in the country and help people refinance their mortgages at today's lower interest rates.

Headlines of the day also influenced the speech.

Obama mentioned North Korea's latest underground nuclear test, which the State Department labeled "provocative" and "extremely regrettable."

With victims of gun violence in the audience at the Capitol, including former U.S. Rep. Gabrielle Giffords, Obama continued his push for tighter gun laws opposed by the influential National Rifle Association and legislators from both parties.

He mentioned 15-year-old Hadiya Pendleton, the Chicago girl killed by gun violence after returning home from taking part in inauguration activities in Washington, saying she was shot a mile from his home in the city.

The girl's parents were guests of first lady Michelle Obama at the address. Also attending was former rock star Ted Nugent, a vocal critic of Obama and any efforts to strengthen gun controls in America.

Obama cited the major provisions of his package of gun proposals, including background checks on all gun sales, a ban on semi-automatic weapons that mimic military weapons, and limiting ammunition magazines to 10 rounds.
